SUMMARY

This retrospective real-world study aimed to describe treatment patterns among Bruton tyrosine kinase inhibitor (BTKi)/B-cell lymphoma 2 inhibitor double-exposed patients with chronic lymphocytic leukemia/small lymphocytic lymphoma, using the Symphony Integrated Dataverse. Among 4,734 double-exposed patients, 1,527 (32.3%) initiated a subsequent line of therapy. The most common immediate subsequent treatments were venetoclax-based regimens (37.6%), covalent BTKi-based regimens (31.3%), and pirtobrutinib-based regimens (19.0%). Treatment selection varied according to the qualifying prior regimen. Median follow-up from initiation of subsequent therapy was 17.4 months (interquartile range [IQR], 7.8-31.1), and median time to treatment discontinuation was 14.6 months (IQR, 6.9-28.2) overall. The estimated 360-day treatment discontinuation rate was 42.2% (95% CI, 39.5-45.1), with higher discontinuation rates observed among patients receiving later lines of therapy (21.9% for second-line, 41.7% for third-line, 44.0% for fourth-line, and 51.0% for fifth-line or later).
